  • Oxidative stress and DNA damage induced by spinosad exposure in

Wenping Xu   Mingjun Yang   Jufang Gao   Yang Zhang   Liming Tao

Spinosad, a neurotoxic insecticide, is widely used for crop protection. In order to elucidate the effects of spinosad on oxidative stress and genotoxicity in Sf9 cells, the levels of lipid peroxidation, the activity of antioxidative enzymes, and DNA damage were measured. The results showed that spinosad caused a time-dependent increase in the formation of malondialdehyde and decrease in the activity of superoxide dismutase and catalase. Further studies confirmed that spinosad induced 8-oxoguanine accumulation in Sf9 cells, which is accompanied by increased expression of DNA repair enzymes (OGG1 and MTH1). The neutral comet assay revealed that spinosad induced significant time-related increases of DNA double-strand breaks in Sf9 cells. Our results indicate that spinosad effectively induced oxidative stress and DNA damage in Sf9 cells.

  • Cardioprotective effects of rutin in rats exposed to pirarubicin toxicity

Ya-Di Wang   Yang Zhang   Bo Sun   Xiao-Wei Leng   Ya-Juan Li   Li-Qun Ren

We established both an acute and chronic cardiac toxicity rat model, which showed pretreatment with rutin attenuated pirarubicin-induced myocardial histopathological injury, electrocardiogram abnormalities, and cardiac dysfunction. Rutin also significantly reduced serum levels of MDA, BNP, CK-MB, CTnT, and LDH and increased serum SOD levels. Treatment with rutin and dexrazoxane resulted in an increase in Bcl-2/Bax ratio (p < 0.05) and reduction in JNK and Caspase-3 protein levels, compared to the pirarubicin group (all p < 0.05). Furthermore, rutin at a dose of 50 mg/kg significantly attenuated the above-mentioned alterations. Our study suggests the antioxidant and anti-apoptotic properties of rutin may be responsible for the cardioprotective effects observed.

  • Methods and network nodes for positioning based on displacement data

Methods, a radio network node (110), a wireless device (120) and a positioning node (130) for obtaining a location estimate are disclosed. The location estimate indicates a location of a target radio node (110, 120). The wireless device (120) is operated to obtain the location estimate by use of functionality provided by a cellular telecommunication system (100). A node (110, 120, 130) refers to any one of the radio network node (110), the wireless device (120) and the positioning node (130). The node (110, 120, 130) obtains (308, 312, 316) a reference point for the target radio node (110, 120). The node (110, 120, 130) obtains (309, 313, 317) displacement information indicating displacement, in relation to the reference point, of the target radio node (110, 120). The node (110, 120, 130) sends (314a, 318a)) the displacement information and/or obtains (310, 314b, 318b) the location estimate by dead reckoning based on the displacement information and the reference point.

  • CD147 blockade as a potential and novel treatment of graft rejection

Jing Luan   Yu Zhao   Yang Zhang   Jinlin Miao   Jia Li   Zhi-Nan Chen   Ping Zhu

Cluster of differentiation (CD)147 is highly involved in the T cell activation process. High CD147 expression is observed on the surfaces of activated T cells, particularly CD4+ T cells. In organ transplantation, it is important to prevent graft rejection resulting from the excessive activation of T cells, particularly CD4+ T cells, which exhibit a key role in amplifying the immune response. The present study aimed to investigate the effects of CD147 blockade in vitro and in vivo and used a transplant rejection system to assess the feasibility of utilizing CD147 antibody-based immunosuppressant drugs for the treatment of graft rejection. The effects of CD147 antibodies were evaluated on lymphocyte proliferation stimulated by phytohemagglutinin or CD3/CD28 magnetic beads and in a one-way mixed lymphocyte reaction (MLR) system in vitro. For the in vivo analysis, an allogeneic skin transplantation mouse model was used. CD147 antibodies were effective against lymphocytes, particularly CD4+T lymphocytes, and were additionally effective in the one-way MLR system. In the allogeneic skin transplantation mouse model, the survival of transplanted skin was extended in the CD147 antibody-treated group. Furthermore, the level of inflammatory cell infiltration in transplanted skin was reduced. CD147 blockade decreased the serum levels of interleukin (IL)-17 and the proportions of peripheral blood CD4+ and CD8+ memory T cells. The data demonstrated that CD147 blockade suppressed skin graft rejection, primarily by suppressing CD4+T and memory T cell proliferation, indicating that CD147 exhibits great potential as a target of immunosuppressant drugs.
