  • Rapid cultivation of aerobic granular sludge in a continuous flow reactor

    Highlights • A novel continuous flow aerobic granular reactor was introduced. • A new cultivation method in continuous flow reactor was proposed. • Mechanism of rapid aerobic granulation was investigated. Abstract Aerobic granular sludge (AGS) was successfully cultivated in a double column cyclic aerobic granular reactor (DCCAGR) within 18 days by inoculating with 30% (w/w) mature aerobic granules and 70% activated sludge, while granulation process and reactor performance was investigated in the continuous flow reactor. The AGS cultivated was white or pale yellow, while granule’s SVI, average particle size, granulation rate, extracellular polymeric substances, polysaccharides/proteins ratio and water content were about 46 mL/g, 1.2 mm, 92%, 37 mg/g MLVSS, 1.4 and 98.3% on the 20th day. Observed from the microscopic structure of the cultivated AGS, a large number of microorganisms tightly adhered together inhibited inside the granules, including coccus, bacillus and protozoa. Finally, effluent chemical oxygen demand, total inorganic chemical demand, ammonium nitrogen and total phosphorus of the system were 33 mg/L, 8.46 mg/L, 7.3 mg/L and 1.26 mg/L, and their removal rates were 97%, 88%, 90% and 87% respectively. Effective hydraulic selection pressure can be created in DCCAGR, while inoculated AGS had positive significance to the agglomeration of other flocculent sludge, which greatly reduced the startup time and consumption of mature aerobic granules.     Aerobic granular sludge which had good performance to pollutants removal was successfully cultivated within 18 days in a pilot scale sequencing batch reactor, about 25% mature aerobic granular sludge was inoculated when the setting time of activated sludge was reduced to 10 min. Anaerobic biological selector was implemented to inhibit filamentous bacteria overgrowth, where the maximum COD could reach to 1703.74 mg/L. The cultivated aerobic granular sludge was irregular and pale yellow, average particle size, SVI, SV30/SV5, PN/PS, EPS and water content were 1.58 mm, 67.64 mL/g, 0.91, 2.17, 268.90 mg EPS/g MLVSS and 98.16% on the 18th day. Mechanism of rapid granulation mainly included crystal nucleus hypothesis and selection pressure hypothesis. The inoculated aerobic granules could maintain stable under short setting time environment, making it directly as the crystal nucleus and the carriers for new particles without obvious disintegration, which eventually shortened the granulation time greatly.   • Tolerance to organic loading rate by aerobic granular sludge in a cyclic aerobic granular reactor

    Sodium acetate as carbon source, tolerance to organic loading rate (OLR) by aerobic granular sludge in a cyclic aerobic granular reactor (CAGR) was investigated by gradually increasing the influent COD. AGS could maintain stability in the continuous flow reactor under OLR <= 15 kg/m(3) d in the former 65 days, and SVI, granulation rate, average particle size and water content was 21 ml/g, 98%, 1.8 mm and 97.2% on the 65th day. However, AGS gradually disintegrated after the 66th day when OLR increased to 18 kg/m(3) d, and granules' properties deteriorated rapidly in a short time. High removal rates to pollutants were achieved by CAGR in the former 65 days, but the removal rates of pollutants dropped sharply from the 66th day. With the increase of OLR and particle size, anaerobic cores inside the granules were formed by massive dead cells, while instability of anaerobic core eventually led to the collapse of the system.     Highlights • A novel rapid cultivation of aerobic granule was introduced. • The feasibility of aerobic granule treating real high C/N ratio chemical wastewater was investigated. • Mechanism of rapid granulation was explored. Abstract Aerobic granular sludge (AGS) was cultivated in a bench scale sequencing batch reactor within 21 days. Strategy of the rapid startup was inoculated with part of mature AGS during cultivation, while aerobic biological selector was implemented for the inhibiting outgrowth of filamentous bacteria and fast selection of zoogloea bacteria. Then, the cultivated AGS was employed for the treatment of solvent recovery raffinate. Stable AGS was successfully domesticated after 55 days under strategy of gradually increase the proportion of real wastewater in influent. The domesticated AGS was orange, irregular shape, smooth and compact. SVI, SV30/SV5, MLVSS/MLSS, EPS, PN/PS, average particle size, granulation rate, (SOUR)H and (SOUR)N of AGS were 19.06 mL/g, 0.97, 0.55, 30.05 mg/g MLVSS, 1.10, 1.28 mm, 98.87%, 32.47 and 7.97 mg O2/h g VSS respectively. Finally, COD, TIN, NH4+–N and TP of the effluent were lower than 25.9 mg/L, 1.64 mg/L, 1.13 mg/L and 0.21 mg/L, and their removal rate was more than 98.43%, 97.12%, 98.02% and 98.09% respectively. Thus, COD, TP removal, nitrification and denitrification were realized in a single bioreactor. The result indicated that the feasibility of AGS for high C/N ratio industrial wastewater treatment.     Inoculated with mature aerobic granular sludge in a sequencing batch reactor, gradually increasing the proportion of municipal sludge deep dewatering filtrate in influent, aerobic granular sludge was domesticated after 84 days and maintained its structure during the operation. The domesticated AGS was yellowish-brown, dense and irregular spherical shape, average size was 1.49 mm, water content and specific density were 98.13% and 1.0114, the SVI and settling velocity were 40 ml/g and 46.5 m/h. After 38 days, NO3-N accumulated obviously in the reactor as lack of carbon sources. When adding 1-3 g solid CH3COONa at 4.5 and 5.5 h of each cycle from the 57th day, the removal rate of TN rose to above 90% after 20 days, where effective COD removal and denitrification were realized in a single bioreactor. Finally, the removal rates of COD, TP, TN and NH4+-N were higher than 95%, 88%, 96% and 99%. (C) 2014 Elsevier Ltd. All rights reserved.
