  • Neuronal seipin knockout facilitates A beta-induced neuroinflammation and neurotoxicity via reduction of PPAR gamma in hippocampus of mouse

    Background: A characteristic phenotype of congenital generalized lipodystrophy 2 (CGL2) that is caused by loss-of-function of seipin gene is mental retardation. Seipin is highly expressed in hippocampal pyramidal cells and astrocytes. Neuronal knockout of seipin in mice (seipin-KO mice) reduces the hippocampal peroxisome proliferator-activated receptor gamma (PPAR gamma) level without the loss of pyramidal cells. The down-regulation of PPAR. has gained increasing attention in neuroinflammation of Alzheimer's disease (AD). Thus, the present study focused on exploring the influence of seipin depletion on beta-amyloid (A beta)-induced neuroinflammation and A beta neurotoxicity. Methods: Adult male seipin-KO mice were treated with a single intracerebroventricular (i.c.v.) injection of A beta(25-35) (1.2 nmol/mouse) or A beta(1-42) (0.1 nmol/mouse), generally a non-neurotoxic dose in wild-type (WT) mice. Spatial cognitive behaviors were assessed by Morris water maze and Y-maze tests, and hippocampal CA1 pyramidal cells and inflammatory responses were examined. Results: The A beta(25-35/1-42) injection in the seipin-KO mice caused approximately 30-35 % death of pyramidal cells and production of Hoechst-positive cells with the impairment of spatial memory. In comparison with the WT mice, the number of astrocytes and microglia in the seipin-KO mice had no significant difference, whereas the levels of IL-6 and TNF-alpha were slightly increased. Similarly, the A beta(25-35/1-42) injection in the seipin-KO mice rather than the WT mice could stimulate the activation of astrocytes or microglia and further elevated the levels of IL-6 and TNF-alpha. Treatment of the seipin-KO mice with the PPAR gamma agonist rosiglitazone (rosi) could prevent A beta(25-35/1-42)-induced neuroinflammation and neurotoxicity, which was blocked by the PPAR gamma antagonist GW9962. In the seipin-KO mice, the level of glycogen synthase kinase-3 beta (GSK3 beta) phosphorylation at Tyr216 was elevated, while at Ser9, it was reduced compared to the WT mice, which were corrected by the rosi treatment but were unaffected by the A beta(25-35) injection. Conclusions: Seipin deficiency in astrocytes increases GSK3 beta activity and levels of IL-6 and TNF-alpha through reducing PPAR., which can facilitate A beta(25-35/1-42)-induced neuroinflammation to cause the death of neuronal cells and cognitive deficits.

  • Novel Mn-based Li-rich layered oxide 0.3Li(2)MnO(3)center dot 0.7LiNi(1/3)Co(1/3)Mn(1/3)O(2) as anode material for lithium-ion batteries

    The possibility of Mn-based Li-rich layered oxides 0.3Li(2)MnO(3)center dot 0.7LiNi(1/3)Co(1/3)Mn(1/3)O(2) as anode material for Li-ion batteries is explored. Ex-situ XRD patterns show that the layered structure remained when discharged to 1.2 V and the phase of Li2O and transition metal (Ni, Mn and Co) emerged when discharged to 0.05 V, indicating that there exists conversion reaction mechanism like transition-metal oxides anode material. The reversible capacities reach 574.2 and 449.8 mAh g(-1) at the current density of 50 mA g(-1) and 200 mA g(-1) respectively after 80 cycles. This study suggests that further optimizing Mn-based Li-rich layered oxides is a promising alternative anode material for Li-ion batteries. (C) 2017 Elsevier B.V. All rights reserved.

  • Simvastatin Enhances Activity and Trafficking of alpha 7 Nicotinic Acetylcholine Receptor in Hippocampal Neurons Through PKC and CaMKII Signaling Pathways

    Simvastatin (SV) enhances glutamate release and synaptic plasticity in hippocampal CA1 region upon activation of alpha 7 nicotinic acetylcholine receptor (alpha 7nAChR). In this study, we examined the effects of SV on the functional activity of alpha 7nAChR on CA1 pyramidal cells using patch-clamp recording and explored the underlying mechanisms. We found that the treatment of hippocampal slices with SV for 2 h induced a dose-dependent increase in the amplitude of ACh-evoked inward currents (I-ACh) and the level of alpha 7nAChR protein on the cell membrane without change in the level of alpha 7nAChR phosphorylation. These SV-induced phenotypes were suppressed by addition of farnesol (FOH) that converts farnesyl pyrophosphate, but not geranylgeraniol. Similarly, the farnesyl transferase inhibitor FTI277 was able to increase the amplitude of I-ACh and enhance the trafficking of alpha 7nAChR. The treatment with SV enhanced phosphorylation of CaMKII and PKC. The SV-enhanced phosphorylation of CaMKII rather than PKC was blocked by FOH, Src inhibitor PP2 or NMDA receptor antagonist MK801 and mimicked by FTI. The SV-enhanced phosphorylation of PKC was sensitive to the IP3R antagonist 2-APB. The SV-increased amplitude of I-ACh was suppressed by PKC inhibitor GF109203X and Go6983, or CaMKII inhibitor KN93. The SV- and FTI-enhanced trafficking of alpha 7nAChR was sensitive to KN93, but not GF109203X or Go6983. The PKC activator PMA increased alpha 7nAChR activity, but had no effect on trafficking of alpha 7nAChR. Collectively, these results indicate that acute treatment with SV enhances the activity and trafficking of alpha 7nAChR by increasing PKC phosphorylation and reducing farnesyl-pyrophosphate to trigger NMDA receptor-mediated CaMKII activation.

  • F-doping and V-defect synergetic effects on Na3V2(PO4)(3)/C composite:A promising cathode with high ionic conductivity for sodium ion batteries

    A facile and simple solid-state reaction route is developed to synthesize F-doping and V-defect Na3V1.98(PO4)(3-x)F-3x/C composites. F-doping is facilitated to decrease the particle size to diminish the pathway of Na+ diffusion. Beneficial by-products are generated due to F-doping and V-defect, Na3PO4 is a typical Na ion conductor and NaVO2 has the layer structure for rapid migration of Na+. The synergetic effect of F-doping and V-defect on the Na+ diffusion is significant. The kinetic behavior is dramatically enhanced and it is beneficial to reinforcing the electrochemical performance. Meanwhile, the extraction of the third Na+ at Na1 site is observed at 4.0 V corresponding to the V4+/V5+ redox couple. The optimized Na3V1.98(PO4)(2.9)F-0.3/C composite delivers an initial charge capacity as high as 143.5 mAh g(-1) and an 116.9 mAh g(-1) discharge capacity at 0.1C. A reversible capacity of 100.6 mAh g(-1) is obtained and it retains 89.3% capacity after 100 cycles at 1C. It presents the highest D-Na(+) (3.66 x 10(-13) cm(2)s(-1)), close to three orders of magnitude higher than the Na3V2(PO4)(3)/C (7.41 x 10(-16) cm(2)s(-1)). Moreover, Ex situ XRD results demonstrate that broadened channels along x and y directions are favorable to improving D-Na(+) and it exhibits the highest D-Na(+) when charging to 3.4 V.

  • Dose-Dependent Neuroprotection and Neurotoxicity of Simvastatin through Reduction of Farnesyl Pyrophosphate in Mice Treated with Intracerebroventricular Injection of A beta(1-42)

    Background: Simvastatin (SV) has been reported to improve dementia and slow progression of Alzheimer's disease (AD), however there are conflicting reports. Objective & Methods: Intracerebroventricular injection of aggregated A beta(1-42) in mice (A beta(1-42)-mice) caused spatial cognitive deficits, long-term potentiation (LTP) impairment, and death of hippocampal pyramidal cells. The present study focused on exploring the dose-dependent effects of SV (10-80 mg/kg) on A beta(1-42)-impaired spatial memory and the underlying mechanisms. Results: The treatment of A beta(1-42)-mice with SV for continuous 15 days could attenuate the spatial cognitive deficits and recover the LTP induction in a "U" type dose-dependent manner. The death of pyramidal cells in A beta(1-42)-mice was significantly reduced by the SV-treatment at 20 mg/kg, but not at dose of 10 or 40 mg/kg, even was aggravated at dose of 80 mg/kg. Hippocampal NMDA receptor (NMDAr) NR2B phosphorylation (phospho-NR2B) was elevated in A beta(1-42)-mice, which was further dose-dependently increased by SV-treatment. Replenishment of isoprenoid farnesyl pyrophosphate (FPP) by applying farnesol (FOH) could abolish the SV-increased phospho-NR2B in A beta(1-42)-mice, but had no effect on the A beta(1-42)-enhanced phospho-NR2B. NMDAr antagonist blocked the neurotoxicity of A beta(1-42) and SV (80 mg/kg) in A beta(1-42)-mice, whereas FOH only inhibited SV (80 mg/kg)neurotoxicity. The SV-treatment in A beta(1-42)-mice corrected the decrease in hippocampal Akt phosphorylation. The PI3K inhibitor abolished the SV (20 mg/kg)-neuroprotection in A beta(1-42)-mice. Conclusion: SV-treatment in A beta(1-42)-mice exerts dose-dependent neuroprotection and neurotoxicity by reducing FPP to enhance the phosphorylation of NR2B and Akt.

  • Panoramic Stereo Sphere Vision

    Conventional stereo vision systems have a small field of view (FOV) which limits their usefulness for certain applications. While panorama vision is able to "see" in all directions of the observation space, scene depth information is missed because of the mapping from 3D reference coordinates to 2D panoramic image. In this paper, we present an innovative vision system which builds by a special combined fish-eye lenses module, and is capable of producing 3D coordinate information from the whole global observation space and acquiring no blind area 360 degrees x360 degrees panoramic image simultaneously just using single vision equipment with one time static shooting. It is called Panoramic Stereo Sphere Vision (PSSV). We proposed the geometric model, mathematic model and parameters calibration method in this paper. Specifically, video surveillance, robotic autonomous navigation, virtual reality, driving assistance, multiple maneuvering target tracking, automatic mapping of environments and attitude estimation are some of the applications which will benefit from PSSV.

  • Research on stereo vision odometry

    The stereo visual odometer in vision based on the navigation system is proposed in the paper. The stereo visual odometer can obtain the motion data to implement the position and attitude estimation of ALV(Autonomous Land Vehicle). Two key technology in the stereo vision odometer are dissertated. The first is using SIFT(Scale Invariant Feature Transform) to extract suitable feature; match points pairs in the feature; and track the feature of fore and after frames of the same point on the object. The second is using matching and tracking to obtain the different 3-D coordinate of the feature of the point on the object; and to compute the motion parameters by motion estimate. The unknown outdoor environment is adopted in the experiment. The results show that the stereo vision odometer is more accurate; and the measurement error dose not increase with the movement distance increasing. It can be used as an important supplement of conventional odometer.

  • An embedded omnidirectional vision navigator for automatic guided vehicles

    Omnidirectional vision appears the definite significance since its advantage of acquiring full 360掳 horizontal field of vision information simultaneously. In this paper; an embedded original omnidirectional vision navigator (EOVN) based on fish-eye lens and embedded technology has been researched. Fish-eye lens is one of the special ways to establish omnidirectional vision. However; it appears with an unavoidable inherent and enormous distortion. A unique integrated navigation method which is conducted on the basis of targets tracking has been proposed. It is composed of multi-target recognition and tracking; distortion rectification; spatial location and navigation control. It is called RTRLN. In order to adapt to the different indoor and outdoor navigation environments; we implant mean-shift and dynamic threshold adjustment into the Particle Filter algorithm to improve the efficiency and robustness of tracking capability. RTRLN has been implanted in an independent development embedded platform. EOVN likes a smart crammer based on COMS+FPGA+DSP. It can guide various vehicles in outdoor environments by tracking the diverse marks hanging in the air. The experiments prove that the EOVN is particularly suitable for the guidance applications which need high requirements on precision and repeatability. The research achievements have a good actual applied inspection.

  • Calibration Research on Fish-eye lens

    A novel calibration method for fisheye lens camera is proposed in this paper. The method is based on equidistance projection model of fisheye lens. The inner structure of the fisheye lens which consists of many pieces of lens is so complex that it is very difficult and unnecessary to construct its physical model. The optical center of the fisheye lens (u0;v0); the virtual focal of the fish-eye lens l and the ration of the distortion in x coordinate Kx and the Ky in y coordinate should be calibrate in the mathematical mode proposed by us. Some special calibration patterns have been designed for calibrating them. Here; the optical center is the precondition for other parameters precise calibration. Therefore; we discuss four different methods to get it. The experiment results show that the methods we present are accurate and stable.

  • Hydrothermal syntheses, crystal structures and properties of two novel 3-D lanthanide(III) coordination polymers constructed with mixed Heterocyclic carboxylic acid

    The hydrothermal reactions of Ln(2)O(3) (Ln = Nd and Eu) with pyridine-2,5-dicarboxylic acid (H(2)pydc) resulted in the formation of two isomorphic three-dimensional (3D) polymeric Ln(III) complexes, [Ln(pydc)(nic) center dot H2O](n) (Ln = Nd(1) and Eu(2)), in which me (nicotinate; also named as pyridine-2-carboxylic acid) might have been formed from the pydc ligands through the C-C bond cleavage and CO2 molecules releasing. Pydc ligands bridge lanthanide centers to form the three-dimensional framework featuring hexagonal channels along the axis a which Lire occupied by bridging me anions and mono-coordinated water molecules. From the topological point of view, two three-dimensional nets are binodal with six- and three-connected nodes, which display a distorted rutile (4.6(2))(2)(4(2).6(10).8(3)) topology. Magnetic measurements (2-300 K) reveal that all polymers possess weak antiferromagnetic property. A strong fluorescence emission spectrum of compound 2 was observed.
