Background: Many stressful conditions, including cardiovascular diseases, induce long-term elevations in circulating catecholamines, thereby leading to changes of the Na/K pump and thus affecting myocardial functions. However, only short-term adrenergic regulation of the Na/K pump has been reported. The present study is the first investigation of long-term adrenergic regulation of the Na/K pump and the potential mechanism. Methods: After acutely isolated Sprague-Dawley rat myocytes were incubated with noradrenaline or isoprenaline for 24 h, Na/K pump high- (I-PH) and low affinity current (I-PL), alpha-isoform mRNA, and alpha-isoform protein were examined using patch clamp, RT-PCR, and Western blotting techniques, respectively. Results: After the short-term incubation, isoprenaline reduced the I-PL through a PKA-dependent pathway that involves alpha(1)-isoform translocation from the membrane to early endosomes, and noradrenaline increased the I-PH through a PKC-dependent pathway that involves alpha(2)-isoform translocation from late endosomes to the membrane. After long-term incubation, isoprenaline increased the I-PL, alpha(1)-isoform mRNA, and alpha(1)-isoform protein, and noradrenaline reduced the I-PH, alpha(2)-isoform mRNA, and alpha(1)-isoform protein through a PKA- or PKC-dependent pathway, respectively. Conclusions: These results suggest that long-term adrenergic Na/K pump regulation is isoform-specific and negatively feeds back on the short-term response. Furthermore, long-term regulation involves transcription and translation of the respective alpha-isoform, whereas short-term regulation involves the translocation of the available alpha-isoform to the plasma membrane. A computer implemented method, data processing system, and computer usable program
code are provided for creating a set of virtual machine image for software. A virtual
software resource template is retrieved. Metadata associated with the virtual
software resource template is copied and the copy of the metadata is modified
to generate personalized metadata for each virtual machine image in the set of
virtual machine images. Then, the set of virtual machine images is deployed using
the personalized metadata.
A systematic method is presented in this paper to show that, based only on the measurements of the input voltage and current, the load impedance of a wireless power transfer system can be instantaneously monitored and load power controlled without using any direct measurement from the load. A new mathematic procedure for deriving the output load information based on input voltage and current is explained. This systematic method, which can be applied to wireless power systems with two or more coils, eliminates the need for sensors and communication devices on the load side, thereby greatly simplifying the power control circuitry. The principle of the load estimation method, the power loss optimization and control scheme are described and favorably verified with measurements obtained from an eight-coil wireless power transfer system.
IL-6 has a dual role in bone remodelling. The ERK1/2 pathway partially upregulated IL-6 secretion in osteocyte-like MLO-Y4 cells exposed to CCF. We have now investigated the possible role of phosphatidylinositol 3-kinase (PI3K)/Akt signalling pathway in the CCF-induced IL-6 expression. MLO-Y4 cells were treated with CCF 2,000 mu strain, 2 Hz, or 10, 30 min, 1, 3 and 6 h. IL-6 expression, Akt and ERK1/2 and PI3K/Akt phosphorylation were determined by RT-PCR, ELISA and Western blotting. Inhibition of PI3K/Akt with LY294002 or ERK1/2 with PD98059 significantly attenuated IL-6 upregulation, and IL-6 expression was abolished by inhibiting both pathways. Inhibition of one pathway downregulated the other's phosphorylation level. In conclusion, concomitant activation of PI3K/Akt and ERK1/2 pathways mediated IL-6 expression in MLO-Y4 cells under CCF.
A new Mg65Cu18Ni6Gd11 bulk metallic glass (BMG) with high level of Ni content, exhibiting glass-forming ability (GFA) of 6 mm, was fabricated by copper mold casting. The compressive strength and plasticity of the Mg65Cu18Ni6Gd11 BMG were 938 MPa and 0.14%, respectively. The composition design of the Mg65Cu18Ni6Gd11: BMG with enhanced GFA had succeeded simply using Gd substituting Y in the Mg65Cu18Ni6Gd11 BMG, which was a result of a comparison on the GFA between the Mg-TM-Gd and Mg-TM-Y BMGs (TM: transition metals). Compared to the Mg-TM-Y BMGs, the Mg-TM-Gd BMGs exhibited improved GFA, which was demonstrated to be mainly attributed to the more negative amorphous formation enthalpy resulting from Gd substituting Y. The effects of rare earth (RE) elements on the GFA of Mg-TM-RE were discussed. This work may suggest an easy way for developing new Mg-based BMGs with improved GFA by rare earth (RE) design.
