  • Different Bla‐g T cell antigens dominate responses in asthma versus rhinitis subjects

    Summary Background and objective The allergenicity of several German cockroach (Bla-g) antigens at the level of IgE responses is well established. However, less is known about the specificity of CD4+ TH responses, and whether differences exist in associated magnitude or cytokine profiles as a function of disease severity. Methods Proteomic and transcriptomic techniques were used to identify novel antigens recognized by allergen-specific T cells. To characterize different TH functionalities of allergen-specific T cells, ELISPOT assays with sets of overlapping peptides covering the sequences of known allergens and novel antigens were employed to measure release of IL-5, IFNγ, IL-10, IL-17 and IL-21. Results Using these techniques, we characterized TH responses in a cohort of adult Bla-g-sensitized subjects, either with (n = 55) or without (n = 17) asthma, and nonsensitized controls (n = 20). T cell responses were detected for ten known Bla-g allergens and an additional ten novel Bla-g antigens, representing in total a 5-fold increase in the number of antigens demonstrated to be targeted by allergen-specific T cells. Responses of sensitized individuals regardless of asthma status were predominantly TH2, but higher in patients with diagnosed asthma. In asthmatic subjects, Bla-g 5, 9 and 11 were immunodominant, while, in contrast, nonasthmatic-sensitized subjects responded mostly to Bla-g 5 and 4 and the novel antigen NBGA5. Conclusions Asthmatic and nonasthmatic cockroach-sensitized individuals exhibit similar TH2-polarized responses. Compared with nonasthmatics, however, asthmatic individuals have responses of higher magnitude and different allergen specificity.

  • High T(g) aliphatic polyesters by the polymerization of spirolactide derivatives

    A series of bicyclic and tricyclic lactide derivatives was prepared using L-lactide as the starting material. Exomethylene-lactide served as a platform for the Die Is Alder addition of various dienes to afford norbornene-, cyclohexadiene-, and isoprene-lactide derivatives. Norbornene-lactide (NL) was also hydrogenated to the saturated norbornane analog. These new lactide derivatives were subjected to ring-opening polymerization using 1,5,7-triazabicyclo[4.4.0]dec-5-ene (TBD) as the catalyst. Homopolymers of poly(cyclohexadiene-lactide) (M(n) = 6.4 kg mol(-1)), poly(isoprene-lactide) (M(n) = 15.3 kg mol(-1)), and poly(norbornane-lactide) (M(n)= 34.4 kg mol(-1)) exhibited glass transition temperatures (T(g)'s) of 119 degrees C, 77 degrees C, and 107 degrees C, respectively. The decomposition temperature (T(d)) of unsaturated PLA derivatives was <= 280 degrees C, whereas saturated poly(norbornane-lactide) has a Td of 300 degrees C. Additionally, a series of poly(norbornene-lactide-s-L-lactide) (P(NL-s-L)) copolymers with varying ratios of NL and L-lactide were prepared. The T(g)'s of the P(NL-s-L) samples ranged from 61 to 100 degrees C, and the values were well-described by the Fox equation.

  • MDM2 309 T/G polymorphism is associated with lung cancer risk among Asians

    Published data on the association between MDM2 309 T/G polymorphism and lung cancer risk are inconclusive. To derive a more precise estimation of the relationship, a meta-analysis was performed. A total of eight studies including 6063 cases and 6678 controls were involved in this meta-analysis. Overall, significantly elevated lung cancer risk was associated with GG variant genotype in recessive model when all the eligible studies were pooled into the meta-analysis (OR = 1.17; 95% CI = 1.02-1.34; P-heterogeneity = 0.06). In the subgroup analysis by ethnicity, significantly increased risks were found among Asians for TG versus TT (OR = 1.20; 95% CI = 1.05-1.37; P-heterogeneity = 0.30), GG versus TT (OR = 1.34; 95% CI = 1.01-1.79; P-heterogeneity = 0.03) and dominant model (OR = 1.26; 95% CI = 1.11-1.43; P-heterogeneity = 0.14). However, no significant associations were found in both Europeans and Africans for all genetic models. This meta-analysis suggests that the MDM2 309G allele is a low-penetrant risk factor for developing lung cancer in Asians. (c) 2009 Elsevier Ltd. All rights reserved.
