  • Combined effects of two antibiotic contaminants on Microcystis aeruginosa

Liu, Ying   Zhang, Jian   Gao, Baoyu   Feng, Suping

Combined toxicity of spiramycin and amoxicillin was tested in Microcystis aeruginosa. The respective 50% effective concentrations (EC50mix) expressed in toxic unit (TU) values were 1.25 and 1.83 for spiramycin and amoxicillin mixed at 1:7 and 1:1, suggesting an antagonistic interaction at the median effect level. Deviations from the prediction of concentration addition (CA) and independent action (IA) models further indicated that combined toxicity of two antibiotics mixed at 1:1 varied from synergism to antagonism with increasing test concentration. Both the EC50mix of 0.86 (in TU value) and the deviation from two models manifested a synergistic interaction between spiramycin and amoxicillin mixed at 7:1. At an environmentally relevant concentration of 800 ng L-1, combined effect of mixed antibiotics on algal growth changed from stimulation to inhibition with the increasing proportion of higher toxic component (spiramycin). Chlorophyll-a content and expression levels of psbA, psaB, and rbcL varied in a similar manner as growth rate, suggesting a correlation between algal growth and photosynthesis under exposure to mixed antibiotics. The stimulation of microcystin-production by mixed antibiotics was related with the elevated expression of mcyB. The mixture of two target antibiotics with low proportion of spiramycin (<50%) could increase the harm of M. aeruginosa to aquatic environments by stimulating algal growth and production and release of microcystin-LR at their current contamination levels. • MULTIPURPOSE NANO-MGO VANADIUM INHIBITOR, METHOD FOR PREPARATION AND USES THEREOF

A method for the preparation of a multipurpose nano-MgO vanadium inhibitor, the nano-MgO vanadium inhibitor thus obtained, and uses thereof; the preparation method uses as raw ingredients a magnesium compound, organic acids, a resin dispersant, an organic solvent and water that are formed by means of a synthesis reaction into oil-soluble nano-MgO. The nano-MgO thus obtained can be used not only as a vanadium inhibitor, but also as an acid neutralizing agent, inhibiting acid corrosion caused by elements such as sulfur and nitrogen during combustion. The method of preparation is simple, and the nano-MgO vanadium inhibitor obtained has a high concentration of Mg, good dispersability and high stability, and can be stored long-term.

  • Hyperphosphorylation Results in Tau Dysfunction in DNA Folding and Protection

Lu, Yang   He, Hai-Jin   Zhou, Jun   Miao, Jun-Ye   Lu, Jing   He, Ying-Ge   Pan, Rong   Liu, Ying

Hyperphosphorylation of tau occurs in preclinical and clinical stages of Alzheimer's disease (AD), and hyperphosphorylated tau is the main constituent of the paired helical filaments in the brains of mild cognitive impairment and AD patients. While most of the work described so far focused on the relationship between hyperphosphorylation of tau and microtubule disassembly as well as axonal transport impairments, both phenomena ultimately leading to cell death, little work has been done to study the correlation between tau hyperphosphorylation and DNA damage. As we showed in this study, tau hyperphosphorylation and DNA damage co-occurred under formaldehyde treatment in N2a cells, indicating that phosphorylated tau (p-Tau) induced by formaldehyde may be involved in DNA impairment. After phosphorylation, the effect of tau in preventing DNA from thermal denaturation was diminished, its ability to accelerate DNA renaturation was lost, and its function in protecting DNA from reactive oxygen species (ROS) attack was impaired. Thus, p-Tau is not only associated with the disassembly of the microtubule system, but also plays a crucial role in DNA impairment. Hyperphosphorylation-mediated dysfunction of tau protein in prevention of DNA structure from damage under the attack of ROS may provide novel insights into the mechanisms underlying tauopathies.

  • Correction of Hair Shaft Defects through Allele-Specific Silencing of Mutant Krt75

Liu, Ying   Snedecor, Elizabeth R.   Zhang, Xu   Xu, Yanfeng   Huang, Lan   Jones, Evan C.   Zhang, Lianfeng   Clark, Richard A.   Roop, Dennis R.   Qin, Chuan   Chen, Jiang

Dominant mutations in keratin genes can cause a number of inheritable skin disorders characterized by intraepidermal blistering, epidermal hyperkeratosis, or abnormalities in skin appendages, such as nail plate dystrophy and structural defects in hair. Allele-specific silencing of mutant keratins through RNA interference is a promising therapeutic approach for suppressing the expression of mutant keratins and related phenotypes in the epidermis. However, its effectiveness on skin appendages remains to be confirmed in vivo. In this study, we developed allele-specific small interfering RNAs capable of selectively suppressing the expression of a mutant Krt75, which causes hair shaft structural defects characterized by the development of blebs along the hair shaft in mice. Hair regenerated from epidermal keratinocyte progenitor cells isolated from mutant Krt75 mouse models reproduced the blebbing phenotype when grafted in vivo. In contrast, mutant cells manipulated with a lentiviral vector expressing mutant Krt75-specific short hairpin RNA (shRNA) persistently suppressed this phenotype. The phenotypic correction was associated with a significant reduction of mutant Krt75 mRNA in the skin grafts. Thus, data obtained from this study demonstrated the feasibility of utilizing RNA interference to achieve durable correction of hair structural phenotypes through allele-specific silencing of mutant keratin genes.

  • PROCESS GROUP AND METHOD FOR ABNORMAL GROUP MEMBER LEAVING PROCESS GROUP

Disclosed are a process group and a method for an abnormal group member leaving a process group. The method includes that: a sending group member implements abnormity checking for group members in the process group to which the sending group member belongs and determines the abnormal group member (S10); the sending group member removes the abnormal group member from the local group member topology (S12); the sending group member sends a forced leaving message so as to notify the abnormal group member to leave the process group after the abnormal group member restores data communication (S14); after receiving the forced leaving message, the abnormal group member leaves the process group (S16). With the present method, the problem, that there is no mechanism to notify the abnormal group member of having been removed from the group after the abnormal group member is removed, is resolved.
