  • KAI1 inhibits lymphangiogenesis and lymphatic metastasis of pancreatic cancer in vivo

Liu, Xu   Guo, Xiao-Zhong   Li, Hong-Yu   Chen, Jiang   Ren, Li-Nan   Wu, Chun-Yan

BACKGROUND: Several studies have shown that KAI1 inhibits tumor metastasis, but its mechanism is not clear. The present study aimed to determine the role of KAI1 in lymphatic metastasis, specifically in pancreatic cancer. METHODS: The KAT1 gene was transfected into the pancreatic cancer cell line MIA PaCa-2 and PANC-1 by using liposomes and selected by G418, and the protein was measured by Western blotting. After successful infection, the cell growth curve was studied by MTT, vascular endothelial growth factor C (VEGF-C) secretion by pancreatic cancer cell were measured by ELISA. The KAI1 and pCMV transfected MIA PaCa-2 cells were renamed as MIA PaCa-2-K and MIA PaCa-2-p. These two kinds of cells were injected into the subcuticular layer of nude mice; both tumor growth and metastasis through the lymphatic nodes were assessed. Lymphangiogenesis in tumors was measured by immunohistochemistry. RESULTS: The VEGF-C secretion was significantly reduced in MIA PaCa-2 cells compared with PANC-1 cells after being transfected with the KAI1. gene. The growth rate of subcutaneous tumors was similar after the injection of MIA PaCa-2-K, MIA PaCa-2, and MIA PaCa-2-p. MIA PaCa-2-K tumors showed slower lymphangiogenesis and lymph node metastasis compared with MIA PaCa-2 and MIA PaCa-2-p tumors. CONCLUSION: The overexpression of KAI1 inhibits the lymphangiogenesis and lymph node metastasis of MIA PaCa-2 pancreatic tumors.

  • CONFIGURATION METHOD AND NODES SUPPORTING MULTI-CARRIER PAGING

The present invention provides a configuration method and nodes supporting multi-carrier paging. The method comprises: a first node receives, on anchor carriers, information about non-anchor carriers which are broadcast by a second node and can be used for paging; the first node selects one carrier from the anchor carriers and the non-anchor carriers, or selects one carrier from the non-anchor carriers; the first node monitors and receives, on the selected carrier, a paging message issued by a mobility management entity by means of the second node. In the case that the first node is a user equipment and the second node is a base station, by configuring, for the user equipment, non-anchor carriers that can be used for transmitting a paging message, and providing a paging carrier selection method for the user equipment, the present invention can lower the limit on paging capacity, and can also enable the user equipment to listen to paging messages on each paging carrier in a relatively balanced manner, so as to reach the purpose of load balancing.

  • Treatment Strategies for Primary Tumors of the Heart in Children: A 10-Year Experience

Liu, Xu   Hong, Haifa   Zhang, Haibo   Xu, Zhiwei   Liu, Jinfen   Qiu, Lisheng

Background The goal of this study was to determine the optimal treatment strategy for children with primary tumors of the heart. Methods We reviewed 88 children with primary heart tumors in our center from January 2004 to December 2013. Operative patients were followed every 6 months in the first postoperative year and then regularly every 12 months; nonoperative patients were followed once a year after diagnosis. Demographic information of imaging, operative details, and postoperative data were obtained from medical records. Statistical analyses were carried out by means of Fisher’s exact tests and Student’s t test. Results Of the 23 patients who underwent surgical removal of the tumors (range, 12 days to 14.4 years; median, 1.3 years), 19 patients had stable postoperative hemodynamics, 4 had low cardiac output, and 2 patients died (mortality, 9%). The follow-up ranged from 6 months to 9 years; 2 patients had tumor recurrence, and 1 patient was lost to follow-up. The rest of the operative patients remained in normal sinus rhythm and had normal cardiac function. The 65 patients treated nonoperatively (range, 1 day to 14.4 years; median, 0.4 years) were followed closely for 1 month to 9.8 years; only 1 patient died (of unknown reasons), 1 patient received a cardiac transplant, and 9 patients were lost to follow-up. Conclusions Operative resection is optimal for primary tumors of the heart in patients with obvious symptoms or hemodynamic changes. Rhabdomyomas have the potential for spontaneous regression, and nonoperative follow-up therefore is acceptable even if symptoms appear. Operative risk is greater in younger patients and in patients with cardiac valvular dysfunction.

  • Noncoding RNAs in human intervertebral disc degeneration: An integrated microarray study

Liu, Xu   Che, Lu   Xie, Yan-Ke   Hu, Qing-Jie   Ma, Chi-Jiao   Pei, Yan-Jun   Wu, Zhi-Gang   Liu, Zhi-Heng   Fan, Li-Ying   Wang, Hai-Qiang

Abstract Accumulating evidence indicates that noncoding RNAs play important roles in a multitude of biological processes. The striking findings of miRNAs (microRNAs) and lncRNAs (long noncoding RNAs) as members of noncoding RNAs open up an exciting era in the studies of gene regulation. More recently, the reports of circRNAs (circular RNAs) add fuel to the noncoding RNAs research. Human intervertebral disc degeneration (IDD) is a main cause of low back pain as a disabling spinal disease. We have addressed the expression profiles if miRNAs, lncRNAs and mRNAs in IDD (Wang et al., J Pathology, 2011 and Wan et al., Arthritis Res Ther, 2014). Furthermore, we thoroughly analysed noncoding RNAs, including miRNAs, lncRNAs and circRNAs in IDD using the very same samples. Here we delineate in detail the contents of the aforementioned microarray analyses. Microarray and sample annotation data were deposited in GEO under accession number GSE67567 as SuperSeries. The integrated analyses of these noncoding RNAs will shed a novel light on coding-noncoding regulatory machinery.
