  • ANTIHYPERTENSIVE EFFECT OF SYNTHETIC TETRANDRINE DERIVATIVES IN SHR RATS

    1. Effects of oral administration of synthetic tetrandrine (TD) derivatives (20 mg/kg per day) for 9 weeks on blood pressure, heart rate, plasma renin concentration (PRC) and vascular reactivities to pressor substances were studied in spontaneously hypertensive (SHR) rats. 2. 7-O-Ethyl fangchinolin (7-O-EFC) and 7-O-isopropyl fangchinolin (7-O-IFC) produced a significant and sustained reduction in blood pressure from the first week of administration. 7-O-EFC reduced heart rate when determined under restraint conditions, but not under unanesthetized, freely moving conditions. 3. TD derivatives produced no effect on PRC. 4. Pressor response to phenylephrine was reduced significantly whereas the response to angiotensin II was enhanced after prolonged administration of 7-O-EFC and 7-O-IFC. 5. These results demonstrate that TD derivatives are potential antihypertensive drugs, and that attenuation of the pressor response to phenylephrine may contribute at least in part of its antihypertensive effect.

  • Expression of annexin A3 in primary cultured parenchymal rat hepatocytes and inhibition of DNA synthesis by suppression of annexin A3 expression using RNA interference

    Annexin A3 is a member of the lipocortin/annexin family, which binds to phospholipids and membranes in a Ca2+-dependent manner. Although annexin A3 has various functions in vitro, its cellular significance is completely unknown. Annexin A3 is not found in rat liver in vivo. In the present study, we investigated the expression of annexin A3 in primary cultured parenchymal rat hepatocytes. Annexin A3 protein was detected in 48-h, but not 2.5-h, cultured hepatocytes using Western blot analysis. The annexin A3 level further increased after an additional 24 h of culture. Annexin A3 mRNA was not detected in 2.5-h cultured hepatocytes but was detected 22 h after the start of culture by RT-PCR analysis, reaching a maximum value after 48 h of culture. To define the role of Annexin A3 in DNA synthesis, RNA interference was used to reduce annexin III gene expression in hepatocytes. The transfection of small interfering RNAs targeting annexin A3 in the hepatocytes reduced the corresponding mRNA and protein expression by approximately 80% and more than 90%, respectively, at 24 h after transfection. In the annexin A3 small interfering RNAs-transfected cells, DNA synthesis, as assessed by [H-3]thymidine incorporation, decreased by approximately 70% not only in the control cultures, but also in the hepatocyte growth factor- or epidermal growth factor-treated cells. These findings show that annexin A3 is expressed in primary cultured parenchymal rat hepatocytes and that the suppression of annexin A3 expression using RNA interference inhibits DNA synthesis.

  • Establishment of spontaneously immortalized rat type 1 astroglial cell lines: The role of p53 in astroglial carcinogenesis

    We established five spontaneously immortalized cell lines using purified rat type 1 astroglia on a rigid transfer schedule. All the cell lines maintained their polygonal shape, regular pavement growth, low saturation density, positive glial fibrillary acidic protein expression, and serum requirements, while none were tumorigenic in nude mice. We then obtained a spontaneously transformed cell line by maintaining the cells for 6 months at a high cell density. Since alterations of the tumor suppressor p53 gene have been reported in the immortalization of some cell lines and in transformation of others, we characterized p53 in immortalized, spontaneously transformed, and 5 N-ethyl-N-nitrosourea (ENU)-transformed cell lines. While each of the ENU-induced or the spontaneously transformed cell lines exhibited p53 gene mutations that resulted in amino acid alterations, no alterations in the p53 gene were observed in any of the immortalized cell lines. Thus, alterations of the p53 protein correlate more strongly with transformation than with immortalization of type 1 astroglia. Immortalization may be regulated by gene(s) other than p53. Spontaneously immortalized type 1 astroglial cell lines may provide a new tool to investigate an initial step of astroglial carcinogenesis. (C) 1996 Wiley-Liss, Inc.

  • A cetylcholine-related bowel dysmotility in homozygous mutant NCX/HOX11L.1-Deficient (NCX-/-) mice - Evidence that acetylcholine is implicated in causing intestinal neuronal dysplasia

    Background/Purpose: Homozygous mutant Ncx/Hox11L.1-deficient (Ncx-/-) mice develop mega-ileo-ceco-colon (mega-ICC) with a caliber change in the proximal colon. The authors investigated the mechanism of intestinal dysmotility in these mice. Methods: Five-week-old Ncx-/- mice with mega ICC were compared with age-matched BDF1 control mice. Jejunum, ileum, and colon were excised from all mice and 1.0-cm-long strips of each organ, each with a resting tension of 0.5g, were suspended in an organ bath filled with Tyrode's solution at 37degreesC and bubbled with a mixture of 95% oxygen and 5% carbon dioxide. Contractile responses to acetylcholine chloride (ACh), histamine, serotonin, and barium chloride (BaCl2) were recorded isometrically. Results: For ACh, Ncx-/- mice had decreased distal colon circular muscle contraction only at lower doses and decreased distal colon longitudinal muscle contraction for all doses compared with controls (P < .05 or P < .01). In the proximal colon, Ncx-/- mice had increased circular muscle contraction only at higher doses and decreased longitudinal muscle contraction only at lower doses compared with controls (P < .01 or P < .05). ACh did not affect jejunum, and there were no significant effects on ileum. There was no response to histamine and serotonin by any part of the bowel, and the response to BaCl2 was the same for both Ncx-/- mice and controls. Conclusions: Only ACh differentially affected muscle contraction in Ncx-/- mice in the proximal and distal colon. Thus, ACh is implicated in causing the bowel dysmotility seen in Ncx-/- mice and human IND. (C) 2004 Elsevier Inc. All rights reserved.

  • OXIDATIVE COUPLING OF METHANE ON PEROVSKITE OXIDES, (BA,CA)(CO,FE)O3-DELTA

    The catalytic activity of perovskite oxides, (Ba,Ca) (Co,Fe)03-delta for oxidative coupling of methane was investigated using temperature-programmed reaction. The addition of a small amount of calcium into BaFeO3-delta resulted in easy oxygen evolution at 200-300-degrees-C. A large amount of carbon dioxide, a small amount of C2 hydrocarbons and a large amount of carbon monoxide were formed at 300, 400 and 700-degrees-C, respectively. On Ba0.95Ca0.05CoyFe1-yO3-delta with y greater-than-or-equal-to 0.8, the production of carbon dioxide was retarded and almost 100% seleCtivitY to C2 hydrocarbons was obtained by the reaction with methane at 550-750-degrees-C. These oxides were found to be prominent catalysts for the oxidative coupling of methane.

  • Effect of surface roughness and calcium phosphate coating on the implant/bone response

    The influence of surface roughness and calcium phosphate (Ca-P) coating on the bone response of titanium implants was investigated. Four types of titanium implants, i.e. as-machined, grit blasted, as-machined with Ca-P sputter coating, and grit blasted with Ca-P sputter coating, were prepared. The Ca-P sputter-coating, produced by using the RF magnetron sputter technique, was rapid heat-treated with infrared radiation at 600 degrees C. These implants were inserted into the left and right femoral condyles and the left and right tibial diaphyses of the rabbits. After implantation periods of 2 and 12 weeks, the bone-implant interface was evaluated histologically and histomorphometrically. Histological evaluation revealed no new bone formation around different implant materials after 2 weeks of implantation. After 12 weeks, bone healing was almost completed. For both tibial and femoral implants, Ca-P coated implants always showed a higher amount of bone contact than either of the non-coated implants. On the other hand, surface roughness improved only the response to implants inserted into the tibial diaphysis. On the basis of these findings, we concluded that 1) deposition of a sputtered Ca-P coating on an implant has a beneficial effect on the bone response to this implant during the healing phase, and 2) besides implant surface conditions the bone response is also determined by local implant site conditions.

  • A patient with acromegaly who showed remarkable improvement of hyperglycemia after treatment with octreotide RID C-8470-2009

    A case with diabetes mellitus associated with growth hormone (GH)-producing pituitary adenoma is described. A 56-year-old woman who had been treated for diabetes mellitus for 3 years, was admitted for the treatment of hyperglycemia. She showed a few acromegalic features and her plasma GH level was 146 +/- 16 ng/ml. After improvement of plasma glucose level by insulin injection, octreotide therapy (100 mu g/8 hours) was started. Seven days after the initiation of octreotide therapy, the fasting plasma glucose level was almost normalized without insulin injection, After the octreotide treatment, urinary C-peptide excretion was significantly decreased and the plasma GH level became within normal range. In this case, octreotide appears to have improved the insulin sensitivity by reducing the plasma GH level.

  • Dehydrogenative cracking of n-butane using double-stage reaction

    Dehydrogenation-cracking double-stage (tandem) reaction of n-butane was studied using a Pt-Sn type dehydrogenation catalyst and a cracking catalyst (rare earth-loaded HZSM-5). n-Butane was firstly dehydroggenated to n-butene (1-and 2-butene) over the Pt-Sn catalyst loaded at the upper part of the reactor. Then n-butene was successively converted to ethylene and propylene over the cracking catalyst loaded at the lower part of the reactor. The yield of light olefins (ethylene + propylene) was 58% at 650 degreesC. The key to obtaining ethylene and propylene in high yield was to determine how the bimolecular reactions of olefins to aromatic and heavier products can be inhibited. It was proved that the loaded rare earths played an important role in inhibiting the bimolecular reactions. (C) 2002 Elsevier Science B.V. All rights reserved.

  • HIGH-RATE OF MIXED GENOTYPES OF HEPATITIS-C VIRUS IN PATIENTS OF AN EPIDEMIC AREA IN JAPAN

    The subjects of this study were 151 patients (69 males and 82 females) who underwent examination and Liver biopsy owing to liver dysfunction in an epidemic area with hepatitis C. Second generation hepatitis C virus antibody (HCV Ab) was positive in 116 (76.8%) of 151 cases. HCV-RNA was detected in 120 (79.5%) by polymerase chain reaction (PCR). In 7 (4.6%) cases, HCV Ab could not be found, but HCV-RNA was detected. Three (2.0%) cases were positive for HCV Ab but negative for HCV-RNA. On the basis of variation in nucleotide sequence within a restricted region in the putative core gene of HCV, HCV genotypes were classed into four types of I, II, III and IV by PCR. The genotypes were identified in 120 cases. Ninety-eight (81.7%) cases carried one of the four types. Type II was found in 76 (63.3%) cases and type III in 22 (18.3%). The other 22 (18.3%) carried simultaneously two different genotypes (mixed type): 21 (17.5%) cases with type II+III and one (0.8%) case with type II+IV. In comparison with the incidence of HCV mixed types in cases with hepatitis C in a non-epidemic area, carriers of mixed types were found at a significantly higher rate in the epidemic area. Liver biopsy of 120 cases with identified HCV genotypes in the epidemic area showed 93 cases of chronic active hepatitis, nine of chronic lobular hepatitis, 10 of chronic persistent hepatitis and eight of liver cirrhosis. No significant correlation could be detected between liver histology and HCV genotypes. Of the 120 cases, 63 (52.5%), 54 (45.0%) and 12 (10.0%) cases had past histories of folk remedies accompanying bleeding, operation and transfusion, respectively. The repetition of these medications may have caused a high ratio of carriers of the mixed genotypes of HCV.

  • Persistent spectral hole burning in Eu(3+)-doped silicate glasses codoping Al(3+) and P(5+) ions

    Eu(3+)-doped SiO(2), Al(2)O(3)-SiO(2), and P(2)O(5)-SiO(2) glasses were prepared by a sol-gel method from metal alkoxides, and their persistent spectral hole burning (PSHB) properties were investigated in relation to the local environment of the EU(3+) ions in glass. Fluorescence line narrowing spectra indicated that EU(3+) clustering occurred in SiO(2) glass. The addition of Al(2)O(3) or P(2)O(5) promoted better dispersion of EU(3+) in the glass matrix, though two sites for EU(3+) ions seemed to exist in Al(2)O(3)-SiO(2) glass. The holes were burned in the 7F(0) - (5)D(0) line of the EU(3+) ions using a Rhodamine 6G dye laser, the hole area of which increased proportionally with the content of hydroxyl groups. The holes that were burned by the photoinduced rearrangement of the OH bonds were thermally refilled and the barrier height of the burnt-state was determined as 0.14, 0.30, and 0.40 eV for P(2)O(5)-SiO(2), SiO(2), and Al(2)O(3)-SiO(2) glass, respectively.
